A huge Chinese container ship passed through the strip of water as thousands of people looked on.
The country's president was among the dignitaries present to celebrate the event.
The enlargement multiplies the canal's capacity by three and is designed to boost trade between the USA and Asia - and divert trade from its competitor the Suez canal.

Friday, the UK took the unprecedented step of voting to leave the
European Union, swiftly followed by the announcement that Prime Minister
David Cameron is to resign in the coming months.
Shortly after the news broke, the pound initially plummeting to a 30-year low, but recuperated some of its losses to sit £1.38 to the dollar by lunch.
Simon Smith, chief economist at foreign exchange broker FX Pro tells Proactive Investors “the reaction of sterling at least is reflected the fact that the world’s not ended, the sun is still shining, the economy is still ticking along and nothing is going to change for at least the next few months”.
He says the pound could be “entering a new paradigm”, or a new era, having trading in a 17.5 figure range, which he adds is “unprecedented”.

Somalia-based terrorist group al-Shabab has claimed responsibility for
an attack on a Mogadishu hotel. The attack killed at least 15 people.
Attackers detonated a suicide car bomb outside the gates of the hotel before storming the building and taking hostages. Somali officials said security forces swept the building Saturday night to end the siege, killing at least four militants.
Hotels frequented by lawmakers and government officials aren't unusual targets for terrorism. Al-Shabab claimed a similar attack that left 16 people dead at the Ambassador Hotel in early June.
The group has been largely driven out of Mogadishu and other Somali cities thanks to intervention from the African Union, but it still has the capability to launch terror attacks.
